  • Making a Difference: Addressing 100 Issues in India and Beyond

    I'm a passionate researcher, on a mission to bridge the gap between academics and the research world. At this moment, through 100 short videos, I aim to tackle 100 critical issues in India and the world, from environmental to educational and socio-economic challenges. My ultimate goal is to make a real impact, not just in awareness but in people's lives, and eventually in the educational sector.

    Project: Computational analysis of d1 receptor modulation by mu-opioid receptor activation in medium spiny neuron (MSN)

    Guide: Prof. Rohit Manchanda, IIT Bombay

    • Accomplished extensive literature review to elucidate the role of endogenous opioids in the reward system.
    • Devised a computational model of the mu-opioid receptors and enkephalin mechanism in the regulation of tonic level and phasic dopamine concentration in MSN.
    • Developed a compartmental model to simulate the biophysical properties of the dendritic spine using Python-NEURON simulation.
    • Modulated D1 receptors and their effect on intrinsic ion channels present on dendritic spines of MSN.
    • Incorporated the dendritic spine model into a complete model of Striatal Neurons using NMODL.

    Project: A study of diagnostic algorithms of Mycobacterium tuberculosis and non-tuberculous mycobacterial disease

    • Cultured 100+ patient samples to diagnose the presence of Tuberculosis.
    • Performed and analyzed the data of microbial staining, GeneXpert System (CBNAAT) and Line probe assay (LPA) to detect M. tuberculosis and multi-drug resistant bacteria.
    • Analyzed Mycobacterial growth (MGIT) and Drug Sensitivity Test (DST) data for low bacterial load samples.
    • Performed different biochemical tests for the identification of Mycobacteria and Nontuberculous Mycobacteria.

    Project: Regulation of histone levels under DNA damaging conditions in the Pathogenic Yeast Candida glabrata

    • Performed sequence analysis of the DNA damage sensor protein kinase RAD53 protein and its gene.
    • Designed cloning and knockout primers for CgRAD53 to delineate its link with Histone H4 level depletion.
    • Attempted to generate CgRAD53 Knockout strain by homologous recombinationmediated gene deletion strategy.

    Project: Cloning, expression and purification of green fluorescence protein (GFP)

    Guide: Prof. Kiran Kondabagil, IIT Bombay

    • Cloned GFP-6xHis gene into E. coli DH5α cells using pET-28a (+) as a vector.
    • Studied the expression of recombinant DNA in E. coli BL 21 after gene transformation using CaCl2.
    • Purified the GFP protein by using Ni-NTA affinity column chromatography.

    Project: Drug designing for targeted lung cancer therapy using bioinformatics tools

    Guide: Prof. Prasenjit Bhaumik, IIT Bombay

    • Sequence alignment of the lung cancer cell surface specific protein Sez6l2 with its homologous superfamily using Clustal X.
    • Generation and evaluation of homology model using I-TASSER and WinCoot.
    • Docking studies of protein with its inhibitor molecule using AutoDock and PyMOL.

    Project: Isolation and biochemical characterization of microorganisms in tap water

    Guide: Prof. Prashant S. Phale, IIT Bombay

    • Isolated and cultured a single bacterial strain from tap water and performed gram staining, capsule staining and spore staining to understand bacterial morphology.
    • Performed the following biochemical tests for bacterial strain identification: IMViC test, Lactose  Fermentation test, Triple Sugar Iron (TSI) test, Sugar utilization test, Nitrate reduction test, Catalase test, Oxidase test, Urease test, Starch hydrolysis test, Casein hydrolysis test and Hemolysis test.
    • Additionally, antibiotic susceptibility test, growth curve determination, Spore Count Assay and T4 Phage enumeration assay were performed.

    Project: A Comparative study between Bovine serum albumin (BSA) and Human serum albumin (HSA)

    Guide: Prof. Dulal Panda and Prof. Samir Maji, IIT Bombay

    • Computationally analyzed Sequence, Hydrophobicity, Secondary structure, and 3D structure of proteins.
    • Analyzed quenching by Acrylamide, Trp fluorescence, and ANS binding using a spectrofluorometer.
    • Studied protein denaturation using heat and GnHCL in far & near UV Circular Dichroism and fluorescence.
